Otome was part of the school's faculty. She is Strict and has extensive martial arts training. If she witnesses Ryoba Aishi commit murder, she will run up and attempt to apprehend her. If she encounters a corpse, she will call the Police and guard the body. If a Teacher's Pet informs her of a body, she will follow them until she sees the corpse, at which point she will act as if she found it herself. If the corpse was moved out of eyesight before she arrives, she will assume it was a prank and get mad at the student.

- Her name, Otome (乙女), means "young lady, little girl, virgin, maiden".
- Her surname, Nagasako, means "long, leader" (naga, 長), while the "sako" part can be written as "hill, uphill path" (嶝) or "urge, force" (迫).
- She teaches all of the rivals except Kaguya Wakaizumi and Chigusa Busujima. She shares the same situation with Shiori Rikitake but she teaches 6 instead of 8.
